Sequencing Verification

Answer the question a sequencing result is meant to answer: does this clone match the intended construct. A read is aligned to the reference, graded on ungapped identity and coverage, and every mismatch behind the grade is listed so the verdict can be checked rather than trusted.

Five real reads against one M13 reference, plus three whole-plasmid pairs that span the verdict — a clean clone that passes, a wrong-vector near miss, and constructs too far apart to call

01

What it does

  1. 01Read to reference alignment with automatic orientation
  2. 02Grade per read, with ungapped identity and coverage stated
  3. 03Coverage bar showing matches, mismatches and gaps along the reference
  4. 04Mismatch list with positions, navigable back to the trace
  5. 05Confirmed regions annotated onto the plasmid map
  6. 06Multiple reads against one reference in a single workspace
02

What it runs on

Alignment and mutation calling run on the cfutils and sangerseqR ports, golden-validated against those libraries. The panel runs on @ports/constructdiff, @ports/verify.

Everything runs client-side. Files are parsed in the browser and projects are saved on this machine, so an unpublished sequence is never uploaded.

Questions

What does the grade mean?
It summarises ungapped identity and coverage of the read against the reference. The underlying numbers and every mismatch are shown next to it, so the grade is auditable.
Can I verify with more than one read?
Yes. Several reads can be compared against one reference in the same workspace, which is what a two-primer confirmation needs.
