Cloning

Recombination Cloning

Run the BP and LR reactions on real sequences and see the entry and expression clones they produce. Recombination is driven by the att sites present in the molecules, so a reaction that cannot proceed is reported instead of assumed.

Real attB fragment, pDONR221 and pET-53-DEST for BP then LR, plus a real floxed plasmid for Cre-lox

01

What it does

  1. 01BP reaction between an attB-flanked fragment and a donor vector
  2. 02LR reaction from an entry clone into a destination vector
  3. 03att site detection in the input molecules
  4. 04Products carried into the project as sequences, ready for the next reaction
  5. 05Chained reactions, so BP then LR runs end to end
02

What it runs on

Recombination runs on the ported cloning engine in packages/ports, exercised against pydna fixtures for the same reactions. The panel runs on @ports/recombination.

Everything runs client-side. Files are parsed in the browser and projects are saved on this machine, so an unpublished sequence is never uploaded.

Questions

Does it handle a full BP then LR chain?
Yes. The entry clone from BP is a sequence in the project, which is what the LR reaction consumes, so the chain runs end to end.
