Gibson Assembly
Give the tool a vector and one or more inserts and it finds the terminal homology between them, orders the fragments, and builds the assembled product. Detected homology is painted onto the map, so a junction that is too short to anneal is visible rather than implied.
Real vector + insert (40 bp homology) for the Gibson Assembly tool
What it does
- 01Terminal homology detection between fragments, with the homology length reported per junction
- 02Product assembly with annotations carried across from the parent fragments
- 03Homology highlighted directly on the plasmid map
- 04Fragments taken from the open project, so a PCR product from another tool can go straight in
- 05Circular and linear products
What it runs on
Assembly runs on the pydna and DnaCauldron ports in packages/ports, golden-tested byte for byte against those Python libraries. The panel runs on @ports/gibson.
Everything runs client-side. Files are parsed in the browser and projects are saved on this machine, so an unpublished sequence is never uploaded.
Questions
- How much terminal homology does it need?
- The homology length is a parameter rather than a fixed constant, and every detected junction reports its own length so a short junction is visible before you build.
- Does the product keep the annotations from the parts?
- Yes. Features on the parent fragments are carried onto the assembled product and remain editable there.
