Circuit construct

The DNA a circuit design emits, drawn to scale at real base coordinates, in the order the library's own placement rules require.

What it does

A circuit design ends in a construct: the sensors, the repressor gate cassettes and the reporter, each a real sequence taken from the characterized library rather than assembled by hand. This draws them along their actual base coordinates, so a cassette three times longer than a promoter looks it.

Two lanes, because they answer different questions. Devices is the architecture — which sensor, which gates, which reporter, and how much of the construct each one occupies. Parts is what a cloning step actually orders: the individual promoters, ribosome binding sites, coding sequences and terminators inside those devices.

What this computes

ReadoutWhat it is
Device spansEach sensor, gate cassette and reporter at its real start and end in the construct, to scale.
Part spansThe individual parts inside each device, each with its type and length.
Device orderThe arrangement the library's placement rules require: which device must come first, which pairs must be adjacent, and which must precede which.
Landing padsThe genomic loci the library integrates into. These are positions in the host genome rather than parts of the construct, so they carry no bases here — but the placement rules position devices relative to them, so they are what the order is anchored on.

How to read it

The order is not cosmetic and it is not the order the gates were listed in. A characterized library declares constraints about where its devices may sit relative to each other and relative to its landing pads, and those constraints exist because arrangement affects behaviour: read-through from one cassette into the next, and the orientation a promoter must face. The order drawn here satisfies all of the constraints that apply to the devices in this design.

Most of a library's placement rules are about devices a given design does not use, so the number of rules that actually bound this construct is much smaller than the number the library declares. Both counts are reported beside the track.

If the constraints cannot all be satisfied at once the panel says so and falls back to listing the devices in the order the logic put them. That is a result, not an arrangement: it is explicitly not the order to build them in.
